From its sites in Cambridge, Mass., and Zurich, Switzerland, Repertoire’s team of more than 120 is creating a new category of immune medicines enabled from its DECODE discovery platform.  The company’s first product in the clinic is a novel multiclonal T cell therapy consisting of T cells derived from the peripheral blood, activated against a curated set of antigens, and armed with a proprietary cytokine.  Repertoire is in clinical-stage immuno-oncology development today, targeting metastatic solid tumors.  In the field of autoimmunity, Repertoire is currently using its proprietary platform to discover novel epitopes and TCRs from patients with type 1 diabetes.  The company is also using its DECODE platform to discover infectious disease antigens and responding T cell clones.

Role Overview

We are seeking a Scientist/Senior Scientist to join the protein engineering and molecular sciences team to support the discovery and development of novel protein therapeutics cross-functionally with the pharmacology and preclinical development team.  The position will focus on the design, generation, optimization, and characterization of protein-based modalities to enable the development of effective therapies for immune-related disorders including autoimmune diseases and cancer.  The Scientist/Senior Scientist will make significant impact on projects and the achievement of group goals predominantly through hands-on experimentation and data analysis.  The candidate will collaborate with multi-disciplinary teams to prioritize hits for protein generation and coordinate biological characterization of molecules.  The ideal candidate will be a motivated, curious, and innovative scientist with a proven ability to solve complex problems.


  • Lead protein engineering efforts in the design of next-generation biologics including management of protein production CRO’s.
  • Characterize target specificity and activity of lead molecules with cell binding and cell signaling assays using flow cytometry.
  • Evaluate affinity and kinetic characterization of lead molecules via immobilized protein assays (e.g. BLI, SPR).
  • Custom ELISA design and development for quantification of lead molecules from serum, plasma, and tissue culture samples.
  • Assess protein quality attributes, such as purity and homogeneity, through assays including SDS-PAGE and SE-HPLC.
  • Development of functional assays to evaluate biological effectiveness of engineered molecules.
  • Independently design and execute experiments, analyze data, and troubleshoot methods.
  • Maintain up-to-date electronic lab notebook.
  • Coordinate with cross-functional teams on the biological characterization of designed molecules; build consensus around candidates and influence decision making through the assessment of data and presentation of results.
  • Communicate experimental design and results to team members and present updates to department.


  • Ph.D. in Biochemistry, Chemistry, Molecular Biology, Structural Biology, or related discipline with 2-8 years post-graduate experience in industry or academia.
  • Detailed knowledge of and experience in protein design and engineering.
  • Must have experience with analysis of protein-protein interactions using BLI or SPR.
  • Well-versed in molecular biology, cloning, subcloning, and construct design with extensive hands-on experience.
  • Experience with custom ELISA generation.
  • Skilled in multi-color flow cytometry.
  • Experience with antibody engineering and/or biophysical characterization.
  • Experience with mammalian protein expression.
  • Extensive experience and track record in protein purification (affinity, IEX, SEC) with demonstrated ability to deliver high-quality, purified protein.
  • Familiarity with or experience in any of the following is a plus:
    • Protein analytical and biophysical techniques including HPLC, DLS, SEC-MALS.
    • Antibody characterization including binding affinity, stability, epitope binning.
    • Multi-specific antibody or bioconjugate generation and characterization.
  • Ability to work in a fast-paced and collaborative environment.
  • Results-oriented, team player.
  • Strong written and oral communication skills.
