From its sites in Cambridge, Mass., and Zurich, Switzerland, Repertoire’s team of more than 120 is creating a new category of immune medicines enabled from its DECODE discovery platform.  The company’s first product in the clinic is a novel multiclonal T cell therapy consisting of T cells derived from the peripheral blood, activated against a curated set of antigens, and armed with a proprietary cytokine.  Repertoire is in clinical-stage immuno-oncology development today, targeting metastatic solid tumors.  In the field of autoimmunity, Repertoire is currently using its proprietary platform to discover novel epitopes and TCRs from patients with type 1 diabetes.  However, we are currently expanding our remit into other autoimmune disorders.  The company is also using its DECODE platform to discover infectious disease antigens and responding T cell clones.

Role Overview

We are seeking a highly motivated Scientist or Senior Scientist/Engineer to work in the tolerance technology platform team, which is responsible for developing and deploying best-in-class immune modulatory therapies based on Repertoire’s proprietary immune decoding technologies.  This is an opportunity to build and characterize a novel immunomodulatory platform.  Working with a dedicated team of scientists and research associates in the tolerance platform team, the successful candidate will take the lead on designing and producing mRNA constructs to deliver antigens, cell modifying payloads, and systemically delivered immunomodulators.  Opportunities to innovate include maximizing expression, antigen processing and immune epitope presentation, silencing mRNA expression in off-target tissues, developing multiplexed strategies for formulation and payload screening, and producing highly immune-silent mRNA constructs.  As part of a multi-disciplinary team, the candidate will work closely with the departments of nanoparticle technology, in vivo pharmacology, protein sciences, antigen discovery and validation, computational sciences and CMC development to advance this new platform through preclinical and clinical development.

The ideal candidate will have relevant experience related to design, production, purification, and molecular and functional characterization of messenger RNA molecules to be used for therapeutic applications as well as interest in developing high throughput in vivo assays for assessing payloads and delivery vehicles.  The successful candidate will be motivated by creating highly active therapeutic molecules, including UTR and ORF design, as well as by establishing a production capability to support ongoing research activities within the tolerance team and across Repertoire.  Working with the broader team, they will drive projects and programs forward from discovery to preclinical to IND enabling studies.  Strong interpersonal, verbal, and written communication skills along with experimental rigor and scientific curiosity are key!  The ability to work in a fast-paced, matrixed team environment, and ability to adapt to evolving priorities are a must.

Key Responsibilities

  • Design and optimize the molecular components of mRNA molecules for antigen expression and presentation in immune cells such as dendritic cells, macrophages, and monocytes as well as in other tissue targets for systemic protein delivery.
  • Collaborate with preclinical biology and disease area teams on characterizing construct designs and payloads for functional activity in various in vitro and in vivo models.
  • Establish, optimize, and innovate processes for in vitro production and purification of mRNA for discovery and therapeutic applications.
  • Contribute to the development of analytical characterization methods for mRNA molecules.
  • Ensure supply of in vivo grade mRNA to the tolerance team and throughout Repertoire.
  • Design and maintain the external supply chain for research grade plasmids for IVT production of mRNA.
  • Analyze, summarize, and communicate team leadership and project teams.
  • Document work in electronic lab notebook and author project summaries as needed.
  • Provide scientific, project, and laboratory leadership to junior scientific staff.  Lead by example with lab safety.


  • B.S., M.S., or Ph.D. in Molecular Biology, Biochemistry, Chemistry, Bioengineering, or a related field.
  • 0-6 years of biotech/pharma experience post Ph.D. or M.S./B.S. plus equivalent experience
    (title will be commensurate to experience).
  • Subject matter expertise in design, production, and analytical characterization of nucleic acid molecules for gene or antigen therapy. 
  • Hands on experience with production and purification of IVT produced mRNA is a plus.
  • Experience with in vitro cell culture and FACs is a plus.
  • Ability to identify, troubleshoot, and solve issues associated with complex materials.
  • Strong organizational skills and excellent attention to detail.
  • Proven track record of being personable, energetic, accountable, and an effective communicator.
  • Must work well independently as well as an integral part of the team.
  • Ability to work in a fast-paced environment.
