Repertoire is seeking a highly motivated and innovative scientist to join our immunotherapy discovery team. Primary responsibility will be the engineering of next-generation biologics and optimization of antibodies and cytokines. This is a unique opportunity to collaborate with colleagues in Discovery and Early Development teams to directly contribute to biotherapeutic discovery campaigns. In-depth understanding of antibody and protein structure-function relationships and in vitro display technologies (e.g. yeast display) will be key to this position. The ideal candidate will be highly proficient in the design of protein/antibody constructs, advanced molecular biology techniques, protein biochemical characterization, and cell biology analyses.
Key Responsibilities Include:
- Integral member of highly collaborative team utilizing yeast display and rational protein engineering to develop novel biotherapeutic modalities for immune therapy
- Apply expertise in protein and antibody structure/function, molecular biology, alternative formats (i.e. conjugates, bispecifics, etc.) ¸ and display technologies (e.g. yeast display) to biotherapeutic discovery and optimization across multiple projects
- Lead execution of protein engineering and antibody discovery/optimization efforts such as humanization, affinity maturation, specificity modulation, and platform improvements
- Perform biophysical characterization such as binding kinetics, protein stability, and specificity; cell-based binding and functional analyses using flow cytometry
- Maintain a timely and accurate laboratory record of all activities
- Present results regularly at team meetings and drive data-driven decisions
Minimum Qualifications:
- PhD in molecular bioengineering, chemical engineering, biochemistry, structural biology, or a related discipline is required with at least 4+ years of post-doctoral training, at least 8 years’ experience (including 4+ years industry experience) required for senior scientist
- Experience with antibody and biotherapeutic discovery and a strong working knowledge of protein and antibody structure/function
- Strong expertise in protein engineering display technologies, preferably yeast surface display, are required
- Must have strong expertise in wide range of molecular biology techniques including molecular cloning, construct design, Gibson assembly and sequence analysis
- High proficiency with protein engineering methodologies, including mutagenesis, library design, and protein display technology (ideally yeast display)
- Protein analytic capabilities, including protein-protein interactions (cellular binding, SPR, Octet), biochemical analyses (affinity chromatography, SEC, IEX, DLS, thermal stability)
- Familiarity with protein modeling tools such as Coot and Rosetta and standard bioinformatic tools (IMGT, Expasy, BLAST)
- Excellent written and oral communication skills
- Strong track record of scientific accomplishments as demonstrated through publications in highly regarded journals and/or intellectual property generation
Preferred Qualifications:
- Antibody structure-function and humanization experience highly desired
- Experience with multicolor flow cytometry is preferred
- Knowledge of immune oncology space is desirable
- Understanding of cytokine/receptor biology and advanced cytokine engineering strategies
- R&D experience in an industry setting with demonstrated successes in biologics drug discovery
